Effect of in vitro High Temperature on Antiviral Function of Interferon and 2' -5' Oligoadenylate Synthetase Activity

Abstract

|
In this report we performed plaque reduction test using Hep-2 cell and vesiculo-stomatitis virus(VSV) at 37¡Éand 39¡Éexamine the effect of high temperature on the antiviral activity of human IFN-¥ã. And we measured the activity of 2-5A synthetase
to
determine the effect of high temperature on the 2-5A system of Hep-2 cells treated with IFN-¥á or IFN-¥ã at 39¡Éand at 37¡É.
In the case of the treatment of Hep-2 cells with IFN-¥á or IFN-¥ã before VSV inoculation, the antiviral actions of IFN-¥á and IFN-¥ãwere greater at 39¡Éthan at 37¡É. In the case of INF-¥áor IFN-¥ã treatment after VSV inoculation, no antiviral
action of
IFN-¥á or IFN-¥ã was boserved. Enhancement of 2-5A synthetase activities by IFN-¥á or IFN-¥ã were greater at 39¡É than at 37¡É.
From this study, it may be concluded that the antiviral action of IFN-¥ãand the activity of the 2-5A system are enhanced by high temperature. It seems that enhancement of the 2-5A system activity at high temperature may contribute to the
enhancement of
antiviral action of INF-¥ã.
It is reported that different characters in 2-5A system activation by IFN-¥áor IFN-¥ã. Thus the mechanisms of enhancement of 2-5A system activation by IFN-¥ãat high temperature should be further studyied.
